Abstract

[ACCESS RESTRICTED TO THE UNIVERSITY OF MISSOURI AT REQUEST OF AUTHOR.] Posttranscriptional gene regulation is an important mechanism of controlling mRNA expression and protein production. The RNA binding protein has been shown to regulate dozens of important mRNA targets that are involved in cell cycling, development, proliferation and survival. Here we investigate the role of HuR in the posttranscriptional regulation of Th2 differentiation and function via its interaction and regulation of the mRNA of the transcription factor GATA-3. We demonstrate that GATA-3 mRNA contains A-U rich elements within its 3'UTR that are required for HuR binding and interaction. Here we also describe a novel model of HuR ablation in lung airway epithelial cells, as well as investigate the role of HuR regulation in T cell activation and T cell Anergy.
