Abstract

[ACCESS RESTRICTED TO THE UNIVERSITY OF MISSOURI-COLUMBIA AT AUTHOR'S REQUEST.] Bioconjugation linkers are used covalently link two molecules, where at least one is a large biomolecule for uses such as tracking cellular function, imaging biomarkers, and disease diagnosis. Generally, a fluorescent molecule is conjugated to the biomolecule of interest as a way to gather data. A new heterobifunctional linker was synthesized to provide turn on fluorescence when attached to a thiol group and hydrazide group. The linker is a coumarin derivative containing a maleimide unit and a reactive aldehyde to form a thioether and hydrozone respectively. Through titrations with N-acetyl-cysteine and acethydrazide sequentially, turn-on fluorescence was observed.
